SAPP, Semi-Automated Protein Production, is a new framework from David Baker:

ecent advances in computational protein design have dramatically outpaced traditional experimental validation methods, creating a critical bottleneck where testing dozens of designs requires multiple weeks of labor-intensive work, lacks standardization for consistent data collection, and prevents systematic large-scale investigations of design parameters.

The researchers developed two complementary workflows: Semi-Automated Protein Production (SAPP) enables rapid production and characterization of hundreds of proteins per day from gene fragments in 48 hours with only 6 hours of bench work, at costs equivalent to a few DNA oligos per construct ($0.07/bp for gene fragments), while the Demultiplexing protocol (DMX) leverages oligonucleotide pools to reduce costs 5-fold further, enabling purification and characterization of over 1,000 designs in arrayed format at $5 per construct.
